Dual Analyte Detection: Free cardiac Troponin I (cTnI) is intrinsically unstable and progressively loses immunoreactivity in solution, while most troponin reference materials contain either Troponin I or Troponin C alone. Spectral’s single-chain Troponin I–C proteins (RP-3500 and RP-3700) covalently link cTnI and cTnC into a single molecule, preserving immunoreactivity, enabling dual antibody recognition with fixed stoichiometry, and providing a more stable and consistent reference material across multiple assay formats [Liu et al., Clin Lab. 2001;47(1–2):19–27. PMID: 11214219].

Product Applications

Calibrators for Immunoassays

RP-3500 and RP-3700 are used to build calibration curves in troponin-based immunoassays. RP-8100 serves the same function in NT-proBNP assays.

Controls for Immunoassays

The same proteins are used as quality control materials alongside calibrators to monitor assay performance across testing runs

Western Blot Standards

RP-3700 at approximately 30 kDa and above 95% purity provides a defined reference band for troponin detection. RP-8100 at approximately 9 kDa per subunit suits low-molecular-weight NT-proBNP detection applications.

What are these proteins used for?

What is the difference between RP-3500 and RP-3700?
Both are single-chain Troponin I-C fusion proteins expressed in E. coli. RP-3500 is the first-generation product: approximately 44 kDa, purity above 85% by SDS-PAGE. RP-3700 is the second-generation product: a refined cTnI fragment (amino acids 28-110) fused to cTnC, approximately 30 kDa, purity above 95% by SDS-PAGE, affinity purified. RP-3700 is the preferred choice where maximum protein purity is required.
